UVEGlyphosate Clean-up Cartridge

Pickering SPE sample cleanup cartridges are small columns containing cation exchange resin used to treat samples prior to injection. The high selectivity and ruggedness of an ion exchange resin provides a simple and universal method for removing matrix interferences from the sample. The Pickering SPE sample cleanup cartridges can be used for the clean up of vegetables, fruits and crop samples in the analysis of Glyphosate and AMPA. These cartridges combine the quality of Pickering Laboratories’ analytical columns with the convenience of small, disposable columns.

Product Description

The cartridges are constructed of high-density polypropylene and hold a standard bed volume of 2 ml of resin. There is a 10 ml reservoir above the resin for the trouble-free addition of sample and extraction solution. A 20_m frit supports the resin packing at the bottom of the cartridge. A convenient snap-off seal leaves a male Luer tip fitting when it is removed. To reduce
or stop the flow use a Luer 3-way stopcock or the yellow end-cap.

SPE sample cleanup cartridges are packed using DI water, and are shipped sealed to prevent the resin packing from drying. Resin may move to the top of the column during transport. This is easily corrected by gently shaking the cartridge and allowing the resin to settle before use.

Cartridge Preparation for Use

Shake cartridge and allow to settle

Remove cap and break off tip

Drain using gravity (do not let it dry out)

Wash 2x with 5mL DI Water

Transfer 1mL of extract to the top of the column.

With time, the sulfonic acid will sometimes leach from the cation exchange resin, which will yield a pink effluent. If this happens, wash the resin with the mobile phase or DI water until the color disappears.

Pickering SPE sample clean-up cartridges are used to treat samples prior to injection. The strong cation-exchange resin is composed of sulfonic acid functional groups attached to a styrene divinylbenzene copolymer lattice. The high selectivity and ruggedness provides a simple and universal method for removing matrix interference from the sample. The SPE sample clean-up cartridges are ideal for the cleanup of vegetables, fruits and crop samples in the analysis of Glyphosate and AMPA.

Sample Preparation

Extraction:

Take 25 g of homogenous sample and add enough water (after estimation of moisture content) to make the total volume of water 125 mL. Blend and Centrifuge.

Matrix Specific Modification:

High water content – reduce sample amount to 12.5 g

High protein content – add 100 μL HCl to 20 mL of extract, shake and centrifuge.

High Fat content – perform the methylene chloride partition twice.

Clean-up

Methylene Chloride Partition:

To 20 mL of aqueous extract add 15 mL methylene chloride. Shake for 2-3 min and centrifuge.

To 4.5 mL of aqueous layer add 0.5 mL acidic modifier solution. Shake and centrifuge.

   

SPE Clean-up:

Prepare SPE cartridge (refer to product abstract PA210)

 Transfer 1 mL portions of SPE mobile phase (refer to method abstract MA206 for formulation)
and discard the effluent.

 Elute analytes with 12 mL SPE mobile phase

   

Concentration:

Evaporate to dryness using rotary evaporator or a vacuum vortex-type evaporator or lyophilize

Re-dissolve in 2 mL of the SPE mobile phase
